When I paired the watch to my Mac (yes you can!), I noticed in System Preferences for the sound input source that the audio input levels did not change when I blew into the directional speaker module.
But when I blew into a small hole on the opposite side, the audio input levels matched exactly.
Therefore the microphone is on the opposite of the speaker module– it makes sense since the speaker fires sound toward the ear while the microphone is facing down toward your mouth.
A close up shows the hole has a protective screen which is probably how water resistance is achieved:
